Introduction
Cereulide, the emetic toxin produced in foods by certain strains of Bacillus cereus, is a heat and acid
stable toxin that causes nausea and vomiting when ingested. In very rare cases, people can die after
ingestion of the toxin. Due to its stability, the toxin may still be present even when B. cereus can no
longer be detected. The presence of cereulide seems to be linked to starch-rich foods like rice (dishes)
and pasta (dishes). However, recent data suggest that the occurrence of food borne outbreaks due to

cereulide is more common to foods in general . The toxin has a cyclic structure and consists of in
total 12 monomers as a repeat of (D-O-Leucine-D-Alanine-L-O-Valine-L-Valine). Several methods
have been developed for the detection and/or quantification of the toxin. Some of these methods are
[3, 4]
nonspecific bio-assays and other methods are specifically based on the chemical analysis using
liquid chromatography with mass spectrometry (LC-MS/MS) for the detection and quantification of the
[5, 6, 7, 8]
toxin . The chemical methods are more specific for cereulide and have, therefore, been chosen as
the starting point for standardization of a method for the quantification of cereulide. Recently, research
has been done for the chemodiversity of cereulide. At least 18 cereulide variants were detected by

UHPLC-TOFMS and ion-trap MS sequencing, among which the previously unknown isocereulides
[10]

1 Scope
This document describes the quantitative analysis of the emetic toxin cereulide using high performance
liquid chromatography (HPLC) or ultra performance liquid chromatography (UHPLC) connected to a
tandem mass spectrometer (LC-MS/MS).
This document is applicable to the analysis of the toxin in products intended for human consumption.

Cereulide is extracted from the food matrix by shaking the sample with acetonitrile. C -Cereulide

is used as an internal standard. The components in the solution are separated using HPLC or UHPLC
and subsequently detected using tandem mass spectrometry (LC-MS/MS). For MS, the electro spray
ionization technique (ESI) is used, using the positive mode. The level of emetic toxin (cereulide) is
expressed as μg cereulide/kg product.
5 Reagents
Use only reagents of recognized analytical grade, unless otherwise specified.
5.1 Water, according to ISO 3696.

NOTE Cereulide (labelled and non-labelled) stock and standard solutions are extremely stable, meaning
over three years when stored in a freezer (6.15)

5.15 Cereulide–standard solution Cer-B, ρ = 100 ng/ml (in acetonitrile).
Pipette (6.10) 500 μl cereulide stock solution A (5.14) in a glass volumetric flask (6.11) of 500 ml, make
up to the mark with acetonitrile (5.2) and mix the solution. Store the solution in the freezer (6.15).

Pipette (6.10) 10 ml cereulide standard solution B (5.15) in a glass volumetric flask (6.11) of 100 ml,
make up to the mark with acetonitrile (5.2) and mix the solution. Store the solution in the freezer (6.15).
5.17 Positive control sample or spiked sample (level approximately 10 ng/g).
6 Apparatus and equipment
6.1 Tandem mass spectrometer, equipped with ESI interface (in positive mode) and multiple
reaction monitoring (MRM) mode.
LC system, pump system (HPLC or UHPLC), degasser, autosampler, column oven.
LC-MS software, suitable of data collection, integration.

6.10 Calibrated plunger pipettes, ranges from 10 μl to 100 μl, 100 μl to 1 000 μl, and 1 000 μl
to 5 000 μl, 2 000 μl to 10 000 μl.
6.11 Glass volumetric flasks, volume of 50 ml, 100 ml and 500 ml according to ISO 1042.
6.12 Glass autosampler vials, with snap/screw cap 2 ml.
6.13 PTFE membrane filters, diameter of 25 mm and 0,45 μm pore size.
6.14 Vortex mixer.
6.15 Freezer, capable of temperatures below −15 °C, preferably below −18 °C.

7 Procedure
7.1 Sample preparation
Store the samples (before and during the experiment) in the freezer (6.15) to prevent growth of micro-
organisms.
Take (100 ± 25) g of a representative part of the sample and transfer it to a sample jar. In case the
amount of sample is limited (for example in case of samples involved in food poisoning), take as much
sample as possible and treat identical as the other samples. In this case, a note should be mentioned
when reporting the result. Homogenize the sample by grinding (6.9).
Weigh 2,5 g, to the nearest mg (6.8), of the homogenized sample into a centrifuge tube (6.6) and
pipette (6.10) 500 μl internal standard solution IS-C (5.12); close the tube with the screw top. Mix
(6.14) about 10 s and let the tubes rest for 30 min. Glass tubes should be used especially when solutions
are stored for a longer time. For short contact times, plastic tubes can be used as well. Add 29,5 ml
acetonitrile (5.2) and close the tube again with the screw top.
Place the tube(s) horizontally on the shaker (6.7) and shake firmly for approximately 1 h. After shaking,
centrifuge the tubes for 10 min at 1 000g to 1 500g (6.3). If the solution is clear without floating
particles, no filtration step is necessary. If not, filter the liquid phase using PTFE membrane filters
(6.13), or transfer 2 ml in a centrifuge tube (6.5) and centrifuge the solution at 10 000g to 12 000g (6.4)
for 10 min.
Fill an auto sampler vial (6.12) and close the vial with a cap. The samples are now ready for analysis.
7.2 Standard preparation
The standards should be prepared directly in the vials. Pipette (6.10) the volumes as specified in

Table 1 with cereulide stock solution Cer-C (5.16), acetonitrile (5.2) (see NOTE) and C -cereulide
6
stock solution IS-D (5.13) into a vial (6.12) and close the vial with a cap. Mix (6.14) the solution for 20 s.
NOTE If additional clean up is needed, heptane extraction may be used to reduce the interfering (fatty)
components. After adding 29,5 ml acetonitrile, add 10,0 ml heptane and close the tubes with a screw top. Place
the tube(s) horizontally on the shaker (6.7) and shake firmly for approximately 1 h. Centrifuge the tubes for 10
min at 1 000g to 1 500g (6.3). Proceed with the sample preparation with the lower acetonitrile layer.

Introduction
Le céreulide, la toxine émétique produite dans les aliments par certaines souches de Bacillus cereus, est
une toxine stable à la chaleur et aux acides qui entraîne des nausées et des vomissements lorsqu’elle est
ingérée. Dans des cas très rares, l’ingestion de la toxine peut entraîner la mort. En raison de sa stabilité,
la toxine peut être toujours présente même lorsque B. cereus ne peut plus être détecté. La présence
de céreulide semble liée aux aliments riches en amidon tels que le riz (et les plats à base de riz) et les
pâtes (et les plats à base de pâtes). Toutefois, des données récentes tendent à indiquer que la survenue
de toxi-infections alimentaires dues au céreulide est plus fréquente dans les aliments d’une manière
[9]
générale . La toxine, de structure cyclique, est constituée de 12 monomères au total, sous la forme
d’une répétition de la séquence (D-O-Leucine-D-Alanine-L-O-Valine-L-Valine). Plusieurs méthodes ont
été développées pour détecter et/ou quantifier la toxine. Certaines de ces méthodes sont des dosages
[3, 4]
biologiques non spécifiques et d’autres sont basées sur l’analyse chimique par chromatographie
[5, 6, 7, 8]
liquide couplée à la spectrométrie de masse (CL-SM/SM) pour détecter et quantifier la toxine .
Les méthodes chimiques étant plus spécifiques pour le céreulide, elles ont été choisies comme point
de départ pour la normalisation d’une méthode de quantification du céreulide. Récemment, des
recherches ont été menées sur la diversité chimique du céreulide. Au moins 18 variants de céreulide ont
n
été détectés par séquençage par SM à piège ionique et CLHUP-TOF-SM, parmi lesquels les isocéreulides
[10]

4 Principe général
Le céreulide est extrait de la matrice alimentaire par agitation de l’échantillon avec de l’acétonitrile. Le

C -céreulide est utilisé en tant qu’étalon interne. Les composants en solution sont séparés par CLHP

ou CLUHP, puis détectés par spectrométrie de masse en tandem (CL-SM/SM). Le spectromètre de masse
est équipé d’une source d’ionisation par électronébulisation (ESI) utilisée en mode positif. Le taux de
toxine émétique (céreulide) est exprimé en µg de céreulide par kg de produit.

5 Réactifs
Sauf spécification contraire, utiliser uniquement des réactifs de qualité analytique reconnue.
5.1 Eau, conformément à l’ISO 3696.
5.2 Acétonitrile, de qualité CL-SM.

5.4 Acide formique, 98 % à 100 %, de qualité analytique.

5.8 Phase mobile A, constituée de 10 mmol/l de formate d’ammonium (5.7) avec 0,1 % (v/v) d’acide

5.9 Phase mobile B, constituée de 0,1 % (v/v) d’acide formique (5.4) dans l’acétonitrile (5.2).

Introduire 10 mg à 0,01 mg près de C -céreulide (5.5) dans une fiole jaugée en verre (6.11) de 100 ml,

dissoudre le produit et ajuster au volume avec du méthanol (5.3). Cette solution n’est pas corrigée en
fonction de la pureté du composé. Conserver la solution au congélateur (6.15).
NOTE Les solutions mères et étalons (marquées et non marquées) de céreulide sont extrêmement stables,
c’est-à-dire pendant plus de trois ans lorsqu’elles sont conservées au congélateur (6.15).

5.14 Solution mère Cer-A de céreulide, ρ = 100 000 ng/ml (dans le méthanol).
Introduire 5 mg à 0,01 mg près de céreulide synthétique (5.6) dans une fiole jaugée en verre (6.11) de
50 ml, dissoudre le produit et ajuster au volume avec du méthanol (5.3). Cette solution n’est pas corrigée
en fonction de la pureté du composé. Conserver la solution au congélateur (6.15).
5.15 Solution étalon Cer-B de céreulide, ρ = 100 ng/ml (dans l’acétonitrile).
Introduire à la pipette (6.10) 500 µl de solution mère A de céreulide (5.14) dans une fiole jaugée en verre
(6.11) de 500 ml. Ajuster au volume avec de l’acétonitrile (5.2) et homogénéiser la solution. Conserver la
solution au congélateur (6.15).
5.16 Solution mère Cer-C de céreulide, ρ = 10 ng/ml (dans l’acétonitrile).
Introduire à la pipette (6.10) 10 ml de solution étalon B de céreulide (5.15) dans une fiole jaugée en verre
(6.11) de 100 ml. Ajuster au volume avec de l’acétonitrile (5.2) et homogénéiser la solution. Conserver la
solution au congélateur (6.15).
5.17 Échantillon témoin positif ou échantillon dopé (niveau 10 ng/g environ).
6 Appareillage et matériel
6.1 Spectromètre de masse en tandem, équipé d’une interface ESI (en mode positif) et du mode de
suivi de réactions multiples (multiple reaction monitoring, ou MRM).
Système CL, système de pompe (CLHP ou CLUHP), dégazeur, passeur d’échantillons, four pour colonne.
Logiciel CL-SM, adapté pour collecter et intégrer les données.

6.10 Pipettes à piston étalonnées, de capacités 10 μl à 100 μl, 100 μl à 1 000 μl et 1 000 μl à 5 000 μl,
2 000 μl à 10 000 μl.
6.11 Fioles jaugées en verre, de volumes 50 ml, 100 ml et 500 ml, etc. conformes à l’ISO 1042.
6.12 Flacons en verre pour passeur d’échantillons, avec bouchon vissé/à pression, 2 ml.
6.13 Filtres à membrane en PTFE, diamètre de 25 mm et taille de pore de 0,45 µm.
6.14 Agitateur vortex.
6.15 Congélateur, pouvant atteindre des températures inférieures à −15 °C, de préférence inférieures
à −18 °C.
7 Mode opératoire
7.1 Préparation de l’échantillon
Conserver les échantillons (avant et pendant l’expérience) au congélateur (6.15) afin d’éviter la
croissance des micro-organismes.
Prélever (100 ± 25) g d’une portion représentative de l’échantillon et les placer dans un flacon à
échantillon. Lorsque la quantité d’échantillon disponible est limitée (par exemple lorsque les échantillons
sont liés à une intoxication alimentaire), prélever autant d’échantillon que possible et procéder comme
pour les autres échantillons. Dans ce cas, il convient d’ajouter une note lors de l’expression du résultat.
Homogénéiser l’échantillon par broyage (6.9).
Introduire 2,5 g, au mg près (6.8), d’échantillon homogénéisé dans un tube à centrifuger (6.6). Ajouter
à la pipette (6.10) 500 μl de solution étalon interne IS-C (5.12) et boucher le tube avec le bouchon à vis.
Mélanger (6.14) pendant environ 10 s puis le laisser reposer pendant 30 min. Il convient d’utiliser des
tubes en verre, en particulier lorsque les solutions sont conservées longtemps. Quand le temps de contact
est réduit, des tubes en plastique peuvent également être employés. Ajouter 29,5 ml d’acétonitrile (5.2)
et fermer de nouveau le tube avec le bouchon vissé.
Placer le ou les tubes horizontalement sur l’agitateur (6.7) et agiter fermement pendant environ 1 h.
Après agitation, centrifuger les tubes pendant 10 min entre 1 000g et 1 500g (6.3). Si la solution est
claire sans particules flottantes, aucune étape de filtration n’est nécessaire. Sinon, filtrer la phase
liquide sur des filtres à membrane en PTFE (6.13), ou transférer 2 ml dans un tube à centrifuger (6.5) et
centrifuger la solution entre 10 000g et 12 000g (6.4) pendant 10 min.
Remplir un flacon pour passeur d’échantillons (6.12) et boucher le flacon. L’échantillon est alors prêt à
être analysé.
7.2 Préparation des étalons
Il convient de préparer les étalons directement dans les flacons. Transférer à la pipette (6.10) les
volumes précisés dans le Tableau 1 de solution mère Cer-C (5.16) de céreulide, d’acétonitrile (5.2)

(voir NOTE) et de solution mère IS-D (5.13) de C -céreulide dans un flacon (6.12). Boucher le flacon.

Mélanger (6.14) la solution pendant 20 s.
NOTE Si une clarification supplémentaire est nécessaire, il est possible d’utiliser une extraction à l’heptane
afin de réduire les composants produisant des interférences (gras). Après avoir ajouté 29,5 ml d’acétonitrile,
ajouter 10,0 ml d’heptane et boucher les tubes avec un bouchon vissé. Placer le ou les tubes horizontalement sur
l’agitateur (6.7) et agiter fermement pendant environ 1 h. Centrifuger les tubes pendant 10 min entre 1 000g et
1 500g (6.3). Poursuivre la préparation de l’échantillon avec la couche d’acétonitrile inférieure.
